We are currently investigating mechanisms of T cell tolerance and Helios is a transcription factor first found to be expressed by Tregs and later on by activated CD4 T cells. This anti-Helios antibody is the only monoclonal Ab commercially available for flow cytometry and we chose the PeCY7 conjugate because we wanted a bright fluorochrome which will work with our current panel for T cells.

Results Summary

Splenocytes were stained with antibodies against CD4 followed by fixation and permeabilization, followed by intracellular staining for Foxp3 and Helios. Result shown is gated on CD4+ cells. We observed that Foxp3+ CD4 T cells expressed high levels of Helios compared to Foxp3- CD4 T cells.
