We wanted to incorporate a second fluorochrome off the UV channel on our LSRII to increase our capacity to 12 colors. We currently use the DAPI channel to detect cell viability using Live/Dead Fixable blue stain and for our second fluorochrome we chose BUV 737.

Results Summary

Splenocytes were stained with antibodies against CD4, CD62L and CD44 BUV 737. A CD44 FMO was included to help with gating. Decent separation between CD44 intermediate vs. CD44 hi T cells.

Anticipate slightly more optimization than your typical antibody. Using the right filter set (730/45) for this fluorochrome is absolutely key to minimizing spill over into the Alexa700 channel.
