The goal was to classify the granulocyte activation status in whole blood of patients with different inflammatory disorders (chronic vs. akute). Therefore, different activation markers were combined in a single panel. This antibody was chosen because the concentration most suitable for this sample material is known.

10 Mins at Room temperature with blocking agent (5µl/100µl)

Secondary Incubation

5µl antibody/100µl whole blood, 30 Mins on ice in the dark

Tertiary Incubation

15 Mins on ice in the dark with BD Blood Lysing Solution, wash 2 times with FACS-Buffer (PBS with 1%BSA)

Results Summary

Good antibody with stable and easily detectable/classifiable signal. No unspecific staining. Be sure to block with Tru Stain Blocking reagent or another blocking agent prior to staining. Be sure to stain with antibody before lysing Erythrocytes
