Draq5 is a membrane permeable dye that stains for DNA. It was used in a 8-color panel to analyze stages of red blood cell development. We detected Draq5 in the PerCP-Cy5.5 channel (detection in the APC channel is another option too). Fluorescence compensation was challenging and titration of Draq5 is highly recommended.

Protocol Overview

Four million cells form the bone marrow were stained with cell-surface antibodies for 30 mins at +4c. Cells were washed and 0.5ul of Draq5 in 100ul of staining media was used to resuspend the pellet. Resuspended cells were analyzed on the flow cytometer within 4 hours.

Tips

Titrate Draq5 for your experimental purpose. Be careful with compensation and voltage adjustments.

Results Summary

Excellent separation between cells that were positive vs. negative for Draq5.
