We use CD34 as a marker gene in our retroviral vector used for the transduction of T cells. We use CD34 because it is not expressed on T cells and we can use reagents to purify our transduced populations and visualize them via flow using CD34 antibody staining. We use these CD34 immuno magnetic beads to purify our T cell populations to only include transduced T cells after the transduction process.

Experimental Design and Results Summary

Application

Purification

Starting Material

Human T cells

Protocol Overview

You incubate your cells with these beads for 30 mins. Wash the cells and run them through a column attached to a magnet. Flow through will be unlabeled cells.

Tips

Don't try too purify too many cells, columns will run slowly.

Results Summary

After CD34 enrichment, there is a definite more pure CD34 population, where the CD34 negative cells have been washed out.

Summary

The Good

High enrichment

The Bad

You lose quite a lot of cells through purification and the yield is always less than calculated.

The Bottom Line

CD34 immuno beads are useful to purify even small CD34 positive populations.
