I have used 3 distinct anti-ninepin antibodies over the past several years. This is by far the best for immunofluorescence. It gives a very bright staining of the centriole subdistal appendages and centriole proximal ends. The staining is also clean.
Immunofluorescence
RPE1 cells, fixed with methanol (-20C)
1:3000 dilution of antibody in PBS with 3% BSA and 0.1% triton
PBS with 3% BSA and 0.1% triton
Green anti rabbit 488 secondary antibody ( Molecular Probes)
DAPI
Fluorescent microscopy
This is an excellent antibody. A tiny amount is sufficient for immunofluorescence on countless samples. Works in human derived cell lines but not in mouse derived cells.
