Protocol Overview

Cells are seeded at high density (1x10^6/ml), and incubated with SB-203580 for 1 hour (20 uM). Cells are then subjected to varying temperatures (40 - 60 degrees C) for 3 minutes, subjected to freeze-thaw cycles to lyse the cells, then snap-frozen. Lysates are evaluated by western blot (anti-p38α, sc-535, Santa Cruz).
