This antibody is used to examine the protein level of total STAT5 in mouse bone marrow cells treated with or without IL-6 and leukemia inhibitory factor for 0.5 h or 1 h. Approximately 10 ug protein was separated on SDS-PAGE gel and transferred to nitrocellulose membrane. After blocking, the membrane was incubated with this antibody @ 1:100 dilution at 4C overnight, followed by HRP-conjugated anti-mouse secondary antibody at room temperature for 1 hour. Proteins were detected with the SuperSignal chemiluminescent western blot detection kit (ThermoFisher Scientific). There was no band revealed at all in membrane probed with this antibody, while membrane probed with the discontinued STAT5 antibody from Santa Cruz (SC-835) showed very clear protein bands.
