Product Name:

Anti-Mouse CD45.2 FITC

Catalog Number:

11-0454-85

One of the studies in our lab is the proliferation of transferred T cells in a donor mouse organ. To do this, we use donor and recipient mice that express different alloantigens of CD45. Thus, we are able to stain for both CD45 molecules and determine which cells are from the donor and which are from the recipient.
We use this antibody for both genotyping out genetic CD45.2 mouse line and for differentiating between CD45.1 and CD45.2 expressing T cells. We selected an antibody with FITC because it isn't very bright, but we expect to have a large number of CD45.2 cells so we want to reduce spillover.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mouse blood

Primary Incubation

Isolate mouse PBMCs and wash before staining 1:200 in FACS buffer for 20 minutes on ice in the dark

Blocking Agent

FACS buffer

Secondary Incubation

None

Tertiary Incubation

None

Detection

LSR II

Results Summary

We have transgenic mice that only express CD45.1 and no CD45.2 on cells. We were able to use this antibody to confirm that these mice only express CD45.2 alloantigen.
