IgE antibodies are integral to allergic reactions. We sought to measure cell-bound IgE on peripheral blood mononuclear cells in healthy and allergic subjects. We used this antibody because it recognizes IgE both in solution and when bound to cellular receptors (i.e., the epitope is not obscured).

Results Summary

Bright staining on basophils (CCR3+), which express the high affinity IgE receptor (FceRI). Minimal background on B cells (CD19+) or T cells (CD3+). Used at the recommended concentration, the antibody produced brighter staining on basophils from allergic subjects (who have higher IgE levels). Used at a 5x lower concentration (1ul), staining was adequate to identify basophils, but showed a reduced ability to distinguish subjects with high vs. low IgE levels.
