Our lab utilizes QIAGEN's DNeasy kit to isolate total DNA from the whole blood of mice. The kit includes all reagents required for the procedure, and the protocol is both simple and time efficient for our lab's needs.

Protocol Overview

Add proteinase K to each blood sample. Add Buffer AL to each sample and incubate for 10 minutes at 56C. Add ethanol to each sample and vortex. Apply each sample to a spin column. Centrifuge, discard the flow through, and complete the wash/spin steps as outlined by the manufacturer (i.e. AW1, AW2). Elute each sample into a clean tube using Buffer AE.

Tips

Perform additional spin to remove all liquid prior to elution. Alter elution incubation time depending on the DNA concentration required/desired.

Results Summary

A NanoDrop can be used to determine concentration and purity of each DNA sample following extraction.

I recommend this product for DNA extraction from blood or tissue. We primarily extract from blood samples, but all of the protocols/applications are outlined nicely by the manufacturer and are also very easy to follow. QIAGEN's products always provide us with great results for our PCR and genotyping experiments.
