This kit is great if you want to isolate DNA and RNA from the same cell samples. In our lab, we isolate both human and mouse cells to characterize their function and gene expression in both healthy human and influenza infected mice. We can then compare gene expression using both RT-PCR and RNAseq.

Starting Material

Single cells isolated from whole tissue

Protocol Overview

Single cells are isolated from whole tissue, added to lysis buffer, and run through a Qiagen Qiashredder column to homogenize the cells. The provided RLT plus buffer is added to the lysate and spun at max speed for 3 minutes. The flow through is then added to the AllPrep DNA spin columns and spun. The column is then used for DNA isolation and the flow through for RNA isolation according to the kit's protocol.

Tips

Don't use RNAse away if you are isolating DNA.

Results Summary

DNA and RNA are isolated from the sample samples. The purity and yield are both good and comparable to other Qiagen kits. We are then able to use the RNA for RNAseq and DNA for RT-PCR.
