Research aim: To measure changes in protein palmitoylation in response to various treatments. Cells were treated with a biotin analog in order to capture palmitoylated proteins. Cell lysates were subjected to SDS-PAGE and probed for biotin incorporation utilizing a streptavidin antibody. The antibody was chosen based on the ease of streptavidin for biotin capture (high affinity). The antibody is conjugated to a flurorpore, therefore, we can avoid the typical primary antibody followed by secondary incubations.

Results Summary

Streptavidin antibody is detectable in a biotin dose dependent manner. Likewise, it does not show up in a loading control with no biotin label. I am really happy with the ease and crisp labeling pattern using this antibody.
