Present study was designed to test the effect of feeding milk on pancreatic beta cell proliferation in pancreas of high cholesterol and fat diet induced diabetic rats.

Starting Material

Rat pancreas tissue section, slides, methanol, acetone, BSA, normal goat serum, primary antibody, Fluorescent labeled secondary antibody, Prolong gold antifade mounting medium.

Protocol Overview

Diabetic rats of 15 weeks age fed with or without specific types of milks were maintained at animal house at uniform nutritional conditions. After treatments, rats were sacrificed and pancreas were collected. Pancreas were fixed in 4% paraformaldehyde and embedded in paraffin wax. Thin sections of 10 micron size were prepared and spread on slides. Unmasking of antigens were done with sodium citrate solution and sections were immunostained for proliferation protein marker. Immunostained sections were mounted in microscopic slides using Prolong gold antifade reagent obtained from life technologies. Mounted slides were examined under fluorescent microscope and images were captured for further analysis.

Results Summary

Very high signals of target proteins were observed in microscope. There was no quenching of fluorescent signals in presence of gold antifad mounting medium even at long exposure of exciting light.
