In the present study, diabetic rats maintained at same nutritional condition, supplemented with or without phytoextract, were tested for pLATS1 levels in muscles using its specific antibodies and normalized with LATS1. For this, after detection of pLATS1 protein levels, nitrocellulose membrane blots were stripped and reprobed with anti-LATS1 antibody obtained from Abcam. Selection of antibody was based on merits of antibodies shown in antibody flyer.

Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:1000)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
2 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

A good intense bands of LATS1 proteins were observed at a molecular weight of 140 KDa. However, there were strong non specific bands at lower molecular weight.
Antibody is specific to LATS1
Non specific band with high signals were also detected
