Experiments were planned to test the effect of specific treatment on differential expression of cell proliferation related proteins in rat muscle tissue. For this, tissues were harvested at different time points of treatments and proteins were extracted using RIPA buffer. An equal amount of proteins from each treatment were resolved in 10% SDS-PAGE, western transferred in nitrocellulose membrane and probed with specific antibody raised against CDC6 protein and blots were normalized with alpha-Tubulin antibody procured from Santacruz Biotech. Selection of antibody was based on previous experience with santacruz biotech antibodies.

Overnight at 4 0C (5% Skimmed milk in TBS with 0.1% Tween 20; dilution 1:1000)
5% Skimmed milk in TBS with 0.1% Tween 20 (for 1hr at RT)
0.5hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:5000)
