We use these beads to purify CD4+ T cells from human PBMCs. The protocol is very straight forward and gives consistent results.

Protocol Overview

Count cells, spin dow. Add 80uL of AutoMACS running buffer/10^7 cells and 20uL of beads/10^10 cells. Incubate at 4C for 15min. Wash cells and resuspend in 3ml. Load the cells on the LS column (also miltenyi), wash 3x with running buffer. Elute the cells using the plunger that comes with the columns.

Tips

Could use a lower amount of beads. I went down to 10uL/10^7 cells and still got a good yield of CD4 cells. You should titer the beads to find the optimal amount for you if you wish to save some beads.

Results Summary

Over 90% pure CD4 T cell population. Good yield. Very little CD4+ cells remaining in the flow through (CD4 negative fraction). Consistent results.
