Our lab routinely uses beads for purification of different population of human cells. We tested this product from BioLegend in efforts to save some money on beads as these are cheaper than competitor's.

Protocol Overview

The protocol we tested was the alternative one using the LS colums (Miltenyi). According to the sales rep this protocol should work just as well as the protocol using the 15ml conical magnet. Resuspend PBMCs at 1*10^8/ml.. Add the nanobeads at 1:10 (10uL/100Ul of cells). Incubate on ice for 15min. Add buffer to 3ml. Put the cells in the LS column, wash 3x, elute cells.

Tips

Consult with the sales rep regarding the protocol. They later told us there are many optimization possibilities we could try.

Results Summary

Poor purity of the CD4+ cell fraction, only 25%.
