Product Name:

Anti-Histone H3 antibody - Nuclear Loading Control and ChIP Grade

Catalog Number:

ab1791

We study epigenetics in embryonic stem cells. We use the H3 antibody as a loading control when analyzing histone modifications after different treatments in mouse ES cells.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Mouse ES cells

Primary Incubation

1:50000 2 hours RT on shaker

Blocking Agent

5% milk

Secondary Incubation

1:3000 in 5% milk for one hour at RT

Tertiary Incubation

N/A

Detection

Exposure with SuperSignal™ West Femto Maximum Sensitivity Substrate, imaged with apparatus from Bio-Rad

Results Summary

This antibody is very, very strong and tends to become overexposed easily in SuperSignal™ West Femto Maximum Sensitivity Substrate if used at the dilution recommended in the manual (with mouse ES cell lysates). To avoid overexposure, I try to use at 1:50000 dilution when working with mouse ES cell samples.

Summary

The Good

Antibody is very strong, can use very little and still pick up signal

The Bad

If too much is used, analyzing resulting image becomes difficult

The Bottom Line

Use at very low concentration and it should work alright!
