I use this gel extraction for purification of Enzymatic reaction ( endonuclease ) and CIP treatment. The quality of plasmid DNA can be directly used for ligation reaction.
Purification of plasmid DNA from enzymatic reaction
Plasmid DNA, endonuclease and CIP
We use 3 times of QiaX buffer, 10ul Silica beads and one volume water for every 250mg of Agarose piece. We followed the rest with the kits' manual for the steps. The beads was washed and dried at RT for 30 min. The plasmid DNA was eluted with pre heated water (55c) to the beads.
Don't overdry the beads
We recovered at least 60-70% plasmid DNA from the extraction.

Nothing
To purify the enzymatic reaction during the cloning step.
