We tried this antibody as a measurement of ER stress in H4IIE rat hepatoma cell line. We treated cells with chemical or physiologic inducers of ER stress such as thapsigargin or the saturated fatty acid, palmitate and then probed for this antibody using western blot.

1:100 in 1:3 Odyssey Block:TBST (0.1% Tween) overnight at 4 degrees C.

Blocking Agent

1:3 Odyssey Block:TBS for one hour at room temperature

Secondary Incubation

1:15,000 Goat-anti rabbit in 1:3 Odyssey Block: TBST (0.1% Tween, 0.02% SDS) for one hour at room temperature

Results Summary

Picked up a whole lot of background. Cannot distinguish what band (if any) is GADD-34. Although I could have increased the antibody dilution to attempt to get rid of the background, I don't think anything would have showed up in the 73 kDA area.
