Product Name:

PathScan RTK signaling antibody array kit (Fluorescent readout)

Catalog Number:

7949

This is a nice kit to detect multiple kinase of the RTK path. It is easy to use and with a good layout.

Application

RTK detection

Starting Material

Tumor cell lines

Protocol Overview

Follow instructions found on the product page at Cell Signaling Technology.

Tips

Use ice-cold PBS to wash your samples, and 500ul of lysis buffer for each 10cm/dish. Perform the assay with 1 mg/ml of lysate. If both slides are being used, it is not recommended to expose them simultaneously in the same development cassette. In this case, leave the second slide in the wash buffer (step 12) while proceeding with steps 13-18 using the first slide. After the first slide is finished, proceed with steps 13-18 using the second slide and freshly diluted LumiGLO®/Peroxide reagent.

Results Summary

Each slide has multiple plot for your controls and the 39 proteins that you want to detect. Each plot intensity has to be quantified using a image analysis software (like Biorad - if you have the chemi-doc).

Summary

The Good

In this kit there are kinase that are really diffucult to find as a single antibody. Really easy to use and the protocol is perfect.

The Bad

Not so cheap for only 16 samples.

The Bottom Line

This kit detects 28 receptor tyrosine kinases and 11 signaling nodes. Nice data layout. Really useful for a preliminary screening or for an RTK overview on your sample before and after treatment.
