Company:

Santa Cruz Biotechnologies, Inc.

Product Name:

YEATS2 Antibody (N-16)

Catalog Number:

sc-103328

Our lab generates a lot of knockouts using Crispr-cas9 technology. We use Western blots and subsequent sequencing to screen the knockout clones. We chose this antibody, because Santa Cruz generally produces good antibodies in our experience.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Whole cell lysates from mouse ES cells

Primary Incubation

1:500 dilution overnight in 5% milk buffer at 4C

Blocking Agent

5% milk

Secondary Incubation

1:3000 antirabbit secondary at RT for 1 hour

Tertiary Incubation

NA

Detection

Exposure with SuperSignal™ West Femto Maximum Sensitivity Substrate, imaged with apparatus from Bio-Rad

Results Summary

There are multiple bands near the 150 kda mark, which makes the WB results hard to interpret. We believe the top band is the specific band, although it is very weak and experiences interference from the much stronger, non-specific band right below it when the membrane is imaged. Not the best antibody for Yeats2.
