The TFF1 ELISA kit from Boster was competitively priced and worked with cell culture media taken from MCF7 cells, a human mammary carcinoma which is well known for overexpression of the TFF1 protein. The kit is very sensitive, detecting TFF1 protein at levels of 10pg/mL. The kit is saturated about 1mg/mL, so in my case, a 1 to 100 dilution of cell culture media was needed in order to be in the linear range of the assay.

Protocol Overview

Allow MCF7 cells to grow in their media for at least 3 days. Dilute media 1 to 100 in sample dilution buffer and add to the wells. Follow the manufacturer protocol from here.

Don't let wells dry during washes

Results Summary

Was able to identify mutants with lower TFF1 protein expression in MCF7 cells.
