Company:

Santa Cruz Biotechnology

Product Name:

GRK 2 Antibody (C-9)

Catalog Number:

sc-13143

The goal of this experiment was to test the endogenous expression level of GRK2 kinase in NIH3T3 cells. GRK2 phosphorylates a wide variety of GPCRs, a process necessary for receptor desensitization. This antibody was chosen because it was widely cited in many published papers.

Experimental Design and Results Summary

Applications

Western Blot

Sample

NIH3T3 cells

Primary Incubation

1:1000 in blocking agent for 16 hr at 4 deg

Blocking Agent

5% milk powder in 1x TBS-T (0.1% Tween)

Secondary Incubation

Donkey anti-mouse IgG HRP used at 1:5000 dilution in blocking buffer for 1 hr at room temp

Tertiary Incubation

NA

Detection

ECL 15 sec

Results Summary

Whole cell extract of NIH3T3 cells was prepared in RIPA lysis buffer and 50 µg of protein was loaded for SDS-PAGE followed by Western Blotting with GRK2 antibody from Santa Cruz Biotechnology. This antibody detects a band at ~75 kDa (the expected size for GRK2).
