Company:

Miltenyi biotec

Product Name:

CD4 Microbeads L3T4 mouse

Catalog Number:

130-049-201

We aim to co-inject bone marrow and CD4 splenocytes into P0 mice to evaluate GVHD response as compared to bone marrow alone. We selected these beads as we have had previous success with isolation kits from Miltenyi.

Experimental Design and Results Summary

Applications

Affinity Purification

Sample

Splenocytes from 2 week old mice

Primary Incubation

15 min at 4 degrees C as instructedy by manufacturer

Blocking Agent

No block used

Secondary Incubation

NA; however we did confirm isolation using anti-CD4 APC from eBioscience; 30 min 4 deg C

Tertiary Incubation

NA

Detection

Flow Cytometry confirmed isolation; FACSCalibur

Results Summary

Final sample was ~93% CD4+ and ~2% CD8+ by FC. We did notice that the sample had fewer cells than expected from previous literature ( ~5% as compared to 20% of splenocyte MNC were CD4+).

Additional Notes

Summary

The Good

Familiarity, ease of use

The Bad

Washing and Titrating steps time consuming

The Bottom Line

Would buy again as you save a step as compared to CD4-fluorphore conjugated beads, but if I wanted to ensure purity I may run through a second time.
