Cells from mice lungs and spleen were incubated with BrdU and whole mycobacterium lysates for 72 hours. Cells were surface stained for CD4 and CD8 and fixed/permeabilized for 15 minutes and stained with anti-BrdU antibodies. Using BrdU staining, it was found that peptide HCL2 resulted in enhanced T cell proliferation in response to Mtb antigens.
