Protocol Overview

Blood was collected from tail vein of diabetic and non-diabetic rats. HbA1c in blood samples was measured by manufacturer’s instructions in the kit. Lysed RBCs were subjected to protease digestion which releases glycated valines from hemoglobin beta chains. These glycated valines then serve as substrates for specific fructosyl valine oxidase (FVO) enzyme. The FVO specifically cleaves N-terminal valines and produces hydrogen peroxide. This, in turn, is measured using a POD catalyzed reaction and a suitable chromagen.
