We routinely perform adoptive transfer of CD4 T cells into B6.Thy1.1 recipients and analyze their proliferation in response to antigen. The purity of isolated CD4 T cells obtained with this kit has been quite variable.

Results Summary

This isolation kit is very simple to use and the steps can be completed in about 30 mins (very short compared to Miltenyi kits that require the samples be run through a column for isolation). However, the purity of the cells has been quite variable from experiment to experiment. My general observation is that better purity is obtained if you are only isolating cells from one mouse. Once you start to pool multiple animals for a big isolation, the purity drops from ~90% to about 78-82%. And no, I did not go over the max limit of cells suggested by the manufacturer.
