The general purpose of my research is to study immune cell populations in the gut in healthy vs disease states. Mast cells are not abundant in healthy intestinal tissue, but can accumulate when the normal state of the intestine is perturbed. The product here was used to assess the abundance and localization of mucosal mast cells in the intestine in the diseased state.

Mouse intestine (pre-fixed, frozen)

Primary Incubation

1:100 in 1% BSA in PBS overnight at 4 degrees

Blocking Agent

5% goat serum in 1% BSA in PBS

Secondary Incubation

Goat anti-rat AlexaFluor594 was used at 1:200 in PBS and was incubated on the section at 1 hour at room temperature in the dark

Results Summary

Mcpt1 is a marker for mucosal mast cells, and seen here, the stain is only observed in the mucosa of the intestine. In healthy tissue, mucosal mast cells are relatively rare, but here, in the tumor of an ApcMin/+ mouse, mucosal mast cells are seen to abundantly infiltrate the tumor.

Additional Notes

It's not shown here, but the stain is absent in mast cell deficient mice, confirming its specificity.
