Transgene expression in vivo is often determined by luciferase expression in combination with luciferin and in vivo imaging software. Luciferase expression is usually presented as Relative Luminescence Units. Through the use of recombinant luciferase, the quantification of RLU to a specific luciferase concentration is possible. This recombinant luciferase was used in vivo to these ends.

Protocol Overview

The recombinant luciferase was dissolved in PBS to concentrations of 50, 5, 0.5 and 0.05 ug doses. These concentrations were injected intramuscularly into naive mice, prior to luciferin administration and in vivo imaging.

Tips

Ensure dose volume remains constant over differing luciferase concentrations.

Results Summary

The log concentrations of recombinant luciferase resulting in differing RLUs following luciferin administration and IVIS imaging provided a standard curve from which RLU values can be converted into ug concnetrations.
