The antibody was used to determine periaxin expression from myelinating cells in mouse tibial nerve cryosections. A control stain on intact sciatic nerve showed correct localization and specificity to peripheral myelin was confirmed by staining of adult mouse corpus callosum (CNS myelin), which was negative.
Immunohistochemistry
Intact adult mouse sciatic nerve, 4% PFA perfusion and 2 hour post fix, cryosectioned. Brain used as negative control.
1:200 dilution into Vector Labs M.O.M. basic kit diluent buffer, over night at 4 degrees C
Vector Labs M.O.M. basic kit blocking reagent, per kit protocol
AlexaFluor 647 goat anti-rabbit IgG at 1:500 in PBS, 1 hour at room temperature

Zeiss LSM 780 confocal microscope
Periaxin was readily detectable and displayed the expected staining pattern with no problem due to background. Corpus callosum did not stain, confirming the specificity of the antibody to periaxin for IHC-IF applications, which is a component of PNS myelin. Image shows staining of longitudinal sciatic nerve section.
Mounted with ProLong Diamond anti-fade with DAPI
Worked well at the dilution used with my normal protocol for sciatic nerve staining. No further optimization needed.
No complaints.
The antibody worked well with the protocol described together with the imaging system used.
