My project involves studying transcriptomic changes that occur during transitional B-cell development in the spleen. Experiments typically involve C57BL/6 mice as well as various mutant strains.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

C57BL/6 splenocytes

Primary Incubation

15-20min in FACS buffer

Blocking Agent

Fc block 5min prior

Secondary Incubation

Detection

Flow cytometry

Results Summary

C57BL/6 mice were injected with Listeria Monocytogenes for 18-48 hours. Splenocytes were stained with various surface markers including activation marker CD69. Increased CD69 was observed after 18 hours but disappeared after 48 hours. Both immature (CD24hi) and mature (CD24lo) B-cells were positive for CD69 after 18 hours.

Summary

The Good

This antibody worked in detecting CD69 after exogenous stimulation

The Bad

The range of detection was limited but detectable

The Bottom Line

This is a good activation marker to use in conjunction with other known B-cell activation markers such as CD86
