This antibody was used to study the role of chemokines on GPCR receptor activation. I used this antibody for an Immunoprecipitation (IP) blot. It is good as I don't see any IgG bands and it gives a single sharp band without any non-specific background. I have selected this antibody because it is mouse monoclonal & my IP was with rabbit polyclonal.

Primary Incubation

Antibody at 1:1000 dilution; at 4 degrees overnight in 5% skim milk powder in 0.1% tween-TBS

Blocking Agent

At RT for 2 hrs in 5% skim milk powder in 0.1% tween-TBS

Secondary Incubation

At RT for 1 hr in 5% skim milk powder in 0.1% tween-TBS & 1:5000 anti-mouse-conjugated with HRP;

Tertiary Incubation

Results Summary

To check the association of CCR4 with Galpha 11 protein. There is no association/dissociation between CCR4/Ga11

Very specific. Doesn't detect any IgG in IP blot

I don't see any drawback but primary dilution is little high

The Bottom Line

The antibody works very good and gives a single specific band. It doesn't detect IgG bands which is good for those interested in doing IP experiments.
