I am conducting research on Parkinson’s disease, looking specifically at how effector T cells are able to modulate and exacerbate the neurodegeneration seen in the substantia nigra of Parkinson’s patients. BV-2 cells are cultured and passaged every 3-4 days in fresh complete DMEM.

Experimental Design and Results Summary

Application

Cell culture

Starting Material

BV-2 cells

Protocol Overview

The BV-2 cells are cultured at roughly 1-2 million cells in a T75 flask in 10 mls of DMEM containing 10% of heat inactivated bovine serum and 2% of supplement. Cells are then re-passaged into a new flask containing the same media every 3-4 days.

Tips

Ensure to use the correct amount of FBS in the medium that your culture requires.

Results Summary

The cells grow rapidly in this supplemented medium and are completely confluent on the plate within 3-4 days.

Summary

The Good

Cells can be cultured at whatever concentration you want so if you need the cells sooner, just plate them in the flask at a higher concentration.

The Bad

If the incorrect amount of FBS is supplemented to the media, the cells will not grow as well.

The Bottom Line

The Life Technologies DMEM media is very high quality and will provide you with a high quality culture of your cells.
