Used for staining of substantia nigra following MPTP intoxication to quantitate surviving dopaminergic neurons. Following incubation of fixed tissues with anti-tyrosine hydroxylase antibody the biotinylated secondary antibody is used to visualize neurons. Tissues are counter-stained with thionin.

Results Summary

Used for quantifying both cell bodies within the SN and termini within the striatum is easy. Optimize the color development step and counter-staining to get the best results so you can clearly see individual bodies and their nuclei. Can easily quantitate neuron numbers with stereology and the striatal density using ImageJ
