This was used to isolate the nuclear extract from the cell lines in order to check the nuclear protein activity in compound treated cells. It was chosen for its ease of isolation and because it is less time consuming.

Protocol Overview

Protocol is followed as per the user guide of the supplier. For more nuclear extract, increase initial cell number. To get more concentrated nuclear extract dissolve it in less (2-4 fold) NER solution. All the steps should be done at 4 degrees C.

Tips

The reagent volume (CER-I, CER-II, NER) needs to be added as per the number of cells used for isolating the nuclear and cytoplamic extract.
