Company:

EMD Millipore

Product Name:

Anti-tyrosine hydroxylase phospho Ser19

Catalog Number:

AB5425

This antibody was used to evaluate the effects that MPTP can have on the phosphorylation status of tyrosine hydroxylase at serine19. Following MPTP, midbrain and striatal regions of the brain were isolated, homogenized, and purified before being loaded onto a gel and transferred to PVDF membrane. Membranes were incubated in primary phospho Ser19 antibody overnight at 4 degrees followed by and HRP-conjugated secondary antibody.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Whole brain homogenate

Primary Incubation

1:1000 in 5% dry milk/PBST. Overnight at 4 degrees.

Blocking Agent

5% dry milk/PBST for 45 - 60 minutes at room temperature

Secondary Incubation

1:10,000 HRP-conjugated secondary, 60 minutes at room temp

Tertiary Incubation

NA

Detection

Femto Chemiluminescent

Results Summary

Data sheet states antibody recognizes protein of 60 kDa which is where I found a dark band. However, there were many other bands at different molecular weights. Could be non-specific binding or differences between performing Western blot on PC-12 cells and brain homogenate. Even though the data sheet states this antibody has only been tested on rats I used it on mice and results were good.

Summary

The Good

Good detection at 60 kDa

The Bad

Also detected bands at other molecular weights

The Bottom Line

This antibody works well for detecting phospho Ser19 from mouse brain.
