I am working on the effects of immunomodulation in Parkinson's disease. Here, splenocytes from T cells are polarized to a Th17 phenotype with the use of several cytokines including TGF-beta1.

Spleens are removed from mice, splenocytes undergo red blood cell lysis and positive selection on a CD4+ column, and are then cultured for 5+ days with a number of different cytokines and antibodies that includes 3ng/ml TGF-beta1
You will be pipetting small volumes of this reagent into your media so make sure to pay careful attention so that all of the reagent actually makes it into the media.
After 5+ days the cells showed a high rate of polarization to Th17 cells
