I am working on the effects of immunomodulation in Parkinson's disease. Here, splenocytes from T cells are polarized to a Th17 phenotype with the use of several cytokines including TGF-beta1.

Experimental Design and Results Summary

Application

Cell work

Starting Material

Splenocytes

Protocol Overview

Spleens are removed from mice, splenocytes undergo red blood cell lysis and positive selection on a CD4+ column, and are then cultured for 5+ days with a number of different cytokines and antibodies that includes 3ng/ml TGF-beta1

Tips

You will be pipetting small volumes of this reagent into your media so make sure to pay careful attention so that all of the reagent actually makes it into the media.

Results Summary

After 5+ days the cells showed a high rate of polarization to Th17 cells

Summary

The Good

Does a good job of polarizing the cells to the correct phenotype

The Bad

Hard to pipet such small volumes of reagent

The Bottom Line

The eBioscience TGFbeta1 works well in the polarization of cells to your desired phenotype in cell culture.
