This antibody was used in a preclinical gene therapy study that investigated the feasibility of single intravenous injections to restore aspartoacylase in a knockout mouse model. It has been published in Molecular Therapy in 2013 [PMID: 23817205 ] Several antibodies were tested in this study including some custom made peptide antibodies as well as some made in a collaborating lab for various applications. This antibody (ab97454) gave the best results in Western blot however its immunofluorescence results were not as great.

Experimental Design and Results Summary

Applications

Western blot

Sample

Brain homogenates

Primary Incubation

Aspartoacylase antibody incubated overnight at 4°C in blocking reagent (1:1000 dilution)

Blocking Agent

LiCor Odyssey Blocking Buffer (PBS) Cat#927-40000

Secondary Incubation

Donkey anti-rabbit IR-conjugated antibody [LiCor cat#926-32213] incubated for 1 hour at room temperature (1:1000 dilution)

Tertiary Incubation

NA

Detection

Odyssey Imager was used to detect and quantify signal

Results Summary

This antibody worked excellently in Western blot analysis of brain homogenates of all animal groups. There was only a slight background staining which is a remarkable improvement over other homegrown antibody batches.
