I performed Confocal Z-stacking in Drosophila embryos expressing mcherry and GFP and stained with DAPI. This microscope allows easy sample preparation and handling. Great quality data with consistency!

DAPI staining for DNA, Embryo collection, de-chorionation and mounting in 50% glycerol with anti-fade agent.

Tips

Use DABCO-antifade mixed with DAPI in 50% glycerol. Make sure the water level is beyond threshhold for auto-ejection while using higher magnification

Results Summary

Great results! Simultaneous imaging for DIC, DAPI, mcherry and GFP. Clear differences in GFP localization in male and female embryos.

Features Summary

No eyepiece, everything is on the monitor, easy to handle and optimize, water immersion lense, temp. controlled chamber.

Easy to use, great quality images

The Bad

Relatively long time to do Z-stack for more than 2 fluors. Also, it is quite expensive !

The Bottom Line

Great piece to have at the core facility routinely performing high resolution confocal microscopy experiments with living tissues and cell lines.
