Our experimental goal was to assess the cleavage of alpha-2 antiplasmin protein by our protease of interest. This antibody was successfully used to measure cleavage by Western blot.
Western blotting
Human alpha-2 antiplasmin protein purified from human plasma (Abcam, catalog #ab77936)
Alpha-2 antiplasmin antibody supplied by Santa Cruz Biotech (catalog #sc-73659). Antibody was diluted 1:1000 in blocking agent and incubated overnight at 4°C.
5% nonfat milk in Tris-buffered Saline with Tween (TBST), incubated 30 minutes at room temperature.
Goat anti-rat IgG-HRP supplied by Sigma-Aldrich (catalog #A9037). Antibody was diluted 1:2000 in blocking agent and incubated for 1 hour at room temperature.

ECL Prime Western Blotting Detection Reagent (GE Amersham, #RPN2232) used following manufacturer instructions.
No background or non-specific bands were detected and signal intensity was generally strong. We have not used this antibody with more complex samples.
1 ug protein can generally be detected on film using exposures of less than 1 minute. Band size is around 70 kDa. Since this product is a monoclonal antibody, it may not detect all cleavage fragments when performing degradation studies.
