Western blot was performed using Anti-IL-10 Monoclonal Antibody (JES3-9D7) (Product # MA5-16749) and a 18 kDa band corresponding to Interleukin-10 was observed in Human rec. IL10 as compared to Human rec. IL6 which was used as a negative control. Mouse rec. IL10 was used to determine species cross reactivity. 250 ng of Human rec. IL10 (Lane 1), Human rec. IL6 (Lane 2), Mouse rec. IL10 (Lane 3) were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel (Product # NP0341BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with F(ab2-Rabbit anti-Rat IgG (H+L Secondary Antibody, HRP (Product # PA1-29927,1:20000 dilution) using the iBright FL 1500 (Product # A44115). Chemiluminescentdetection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
