Description

Description: This assay employs a two-site sandwich ELISA to quantitate GTPBP2 in samples. An antibody specific for GTPBP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GTPBP2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GTPBP2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GTPBP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: The deduced protein contains 514 amino acids. Phylogenetic analysis of G protein family members suggested that GTPBL2 shares homology with a distinct cluster of G proteins that includes human, mouse, and C. elegans GTPBP1 and mouse Gtpbp2. Northern blot analysis revealed a major 2.9-kb transcript abundantly expressed in heart, placenta, skeletal muscle, pancreas, and testis; expression was lower in brain, lung, kidney, and ovary.
The mouse and human proteins share 98% amino acid identity. Northern blot analysis of mouse tissues revealed a different pattern of expression than that found in human tissues, with highest expression in thymus, spleen, and testis, and lower expression in brain, heart, lung, liver, kidney, and skeletal muscle
