 Taq Polymerases

Taq polymerase is a DNA polymerase named after the thermophilic bacterium Thermus aquaticus from which it was originally isolated. The thermaphilic bacterium thrives in the hot environments surrounding underwater volcanic vents and consequently, its enzymes are thermostable. The thermostability of these enzymes is the critical feature that allowed the development of PCR, one of the key inventions in molecular biology which has radically changed how molecular research is conducted. This technique is used to amplify DNA utilizing the amazing processivity of the Taq DNA polymerase. When researching which Taq polymerase to purchase for your own experiments, there are a large number of factors to consider and the choice you make will be driven to a large extent by what you wish to accomplish. Factors you might wish to consider include: the yield (i.e. processivity of the Taq), the fidelity or error rate of the Taq, whether you want a Taq which will amplify long sequences of DNA or whether you need to amplify through regions of DNA which are complex or GC rich. These may be some of the more commonly considered factors but the list is by no means exhaustive. For example, Taq is also available with added buffers and/or dye for subsequent gel electrophoresis or in a kit with different buffers for optimization of your PCR reaction. Finally, Taq for hot-start PCR is available to reduce non-specific primer extension. Mulitple cocktails of different Taq enzymes and buffers are available on the market today.
