Luciferase Assay Kits

Many laboratories these days use luciferase as a reporter enzyme. There are a wide variety of luciferase assay systems available, and a selection of these is listed below. When choosing a kit, there are a number of important issues to consider. Firstly, kits are designed for a particular luciferase (for example firefly or sea pansy), so check your expression system before buying. Some kits are also designed with a specific expression system in mind – bacteria, mammalian or plant cells. Take a careful look at the specifications of individual kits, some may be adversely affected by tissue culture reagents such as phenol red or serum supplements. Take note also of the stability and storage conditions of the reagents. Luciferase kits often have a short shelf life. Another difference between kits is in the protocols. Kits that require fewer steps are normally homogenous, meaning that a cell lysis buffer and luciferase substrate are added together. Non-homogenous kits are also available with a separate lysis buffer and substrate mix which can be useful if you wish to transfer a sample from one plate to another for analysis. Check the kit components carefully as the lysis buffer may need to be purchased separately. Check whether the buffer will enable samples to be frozen and analyzed at a later date; this can be very useful for large scale assays or making positive and negative controls. Another issue to consider is the format of each kit. If planning to do high-throughput assays in 96-well plates, take a look at kits that are designed for extended signal stability; this prevents signal being lost over the time it takes to read a whole 96-well plate.
