Synthesis of custom oligonucleotides is a procedure now outsourced by most laboratories. Oligonucleotides have a variety of uses from primers in PCR reactions to probes in microarray and other types of hybridization experiments. Companies offering custom oligo synthesis services offer an array of options. When choosing a company, you must first determine exactly what type of oligonucleotide you require, how much and how pure. Companies usually have fixed minimum and maximum lengths and offer options of scale (usually from nmole to µmole concentrations) as well as different purification methods. In some cases, oligos are available prepacked into 96-well plates for high-throughput experiments. Most companies also offer labeled or blocked oligos for specific applications. The second group of factors you must consider is the overall service provided by the company. Prices for standard oligonucleotides are usually calculated per base pair. It may be worthwhile collaborating with other research groups or institutions before approaching companies: some may be able to find better prices for groups with lots of ordering power. Also consider the location and size of the manufacturing facilities as well as their delivery schedule: the best oligo synthesis services should be both fast and reliable. The customer-company interface is extremely important: ordering online or by email is often fastest. Finally, of course, quality control measures and sequence guarantees are also important.
