 Maximum DNA Yield for Minimum Fuss

Genomic DNA is a valuable source of information in many research projects. Genomic DNA Isolation Kits such as those shown below, are available that make isolating the DNA simple. In general, these kits achieve genomic DNA isolation by breaking down the tissue and lysing the cells, removing cellular material (membranes, proteins and RNA) before extracting the DNA by binding to a column, extracting with organic solvents or and/an alcohol precipitation step. When choosing a kit, there are a number of factors to consider. A good place to start is the source or sources of your genomic DNA. Some kits are able to extract DNA from a wide variety of tissues such as blood, hair follicles, plant, bacterial or animal cells with minor changes in protocol. Others are designed for a particular tissue type, and in some cases species-specific kits are available. Secondly, consider the size of your samples, and the amount of DNA you wish to extract - kits can be restricted to a certain sample size or volume because of the limits of the columns or reagents used. It is worthwhile having a close look at individual protocols as there are a number of different ways to lyse cells, remove proteins and RNA and total processing time can vary. Take note of how the DNA is extracted – some kits use organic solvents which have safety implications and other kits may use an alcohol precipitation or wash steps, which can affect future DNA manipulations if not removed completely. Also take a moment to consider the number of samples you wish to process and frequency of kit use –some kits may be more suitable for managing multiple samples at once. High-throughput specific kits are also available.
