Peptides are synthesized by coupling the carboxyl group or C-terminus of one amino acid to the amino group or N-terminus of another; this can be done in liquid or solid phase by automated synthesizers. Synthetic peptides are extensively used in research for structure-function analysis, binding assays, receptor agonist/antagonists or as immunogens for the production of specific antibodies. Some very important aspects are the quality and length limit of the synthesized peptides, thus it is important to consider the quality control (QC) methods and the availability of the QC report when choosing a peptide synthesis service. QC is usually done by mass spectrometry and analytical HPLC. In addition, amino acid sequencing and/or analysis can often be requested. Other important features one should compare are the flexibility of company regarding the quantity of the peptide they synthesize, whether they can synthesize linear or cyclic peptides of various lengths as well as the availability of modifications such as: acetylation, amidation, biotinylation, phosphorylation, disulfide bridge formation (cyclization), fluorescein, myristylation, phosphorylation, rhodamine, and non-standard residue incorporation, etc. When above criteria are equally met by companies additional criteria such as price, fast delivery, and ease of ordering (online, by phone, fax and email) should be considered. To obtain the best experimental results for your application, it is also important to choose a company that offers technical support from the design phase of your peptide to the right choice of modifications, as well as troubleshooting throughout the process.
