Find Your Phage

In peptide display, a peptide is expressed as a fusion with a native phage protein. The strength of the system lies in the fact that when the phage displaying the peptide of interest has been identified, the DNA encoding that peptide resides within the phage. In general, researchers who are working with phage display systems are looking for a peptide which interacts with a specific target protein. Therefore, a critical step in working with any peptide display system is the selection of the phage expressing the peptides which specifically bind to the target protein. This selection process is generally performed via a technique called biopanning. Biopanning is performed by mixing the peptide-displaying phage with the target protein, washing, and then eluting the bound phage. In most cases, the target protein is fixed to a solid support, usually a 96-well plate. However, fixing the target protein to a solid support may inactivate or block the site to which the displayed peptide would react. In this case, the binding should be done in solution. If the bait protein you are working with is particularly sensitive to denaturation or sufficiently small that it is likely to have its binding site obscured when fixed to the well of a plate, you will want to consider a biopanning kit with the option of biopanning in solution. Be warned, however, that biopanning on a solid-support is much simpler so, if possible, use a 96-well plate.
